Optimalisasi Metode Ekstraksi DNA Daun, Kulit Kayu dan Kayu Pinus merkusii

Abstrak. Keberhasilan ekstraksi DNA sangatlah penting dalam proses investigasi genetika molekuler. Penggunaan sampel biasanya berupa organ tanaman muda karena diketahui dapat menghasilkan ekstrak yang bagus. Kayu dan kulit kayu juga berpotensi sebagai sumber sifatnya mudah didapat. Namun pengujian efektifitasnya untuk spesies Pinus merkusii belum pernah dilaporkan. Tujuan penelitian membandingkan hasil dari tiga jenis menggunakan metode yaitu CTAB, CTAB dengan mercaptoethanol DNeasy Plant Mini Kit. Hasil dikuantifikasi alat nanophotometer divisualisasi gel agarosa. Dari ketiga DNA, memperoleh kemurnian lebih bagus baik pada uji kuantitas maupun kualitas. Semua nilai optical density ratios di luar rentang (λ260/280 = 1,8 – 2,0 ng/µl λ260/230 ng/µl) kayu, daun khusus Kit mencapai 1,8-2,0 mercaptoethanol. tidak menunjukkan pita diinterpretasikan sama sekali, sedangkan KK3 D3 lainnya diinterpretasikan.Kata Kunci: Ekstraksi, KitAbstract.
